Evaluation of Central Nervous System depressant action of catharanthus roseus G. Don leaves in mice

Abstract

Catharanthus roseus leaves were investigated to assess the CNS depressant activity by using locomotor activity and pentobarbitone induced sleeping time in experimental animal models. Successively extracted petroleum ether, chloroform and ethanol extracts were used in the experiment. Petroleum ether extracts showed significant (p<0.001) and dose dependant CNS depressant activity at the dose of 200 and 400mg/kg oral administration of extracts. Diazepam 4mg/kg, i.p. was used as standard drug. The dose of 100mg/kg didn't show significant result in the experimental animals. Petroleum ether extract showed presence of steroids, tannins, flavonoids, glycoside, alkaloids and phenolic compounds in qualitative chemical analysis. Petroleum ether extracts is rich in steroid and phenol content including flavonoids, which are reported responsive chemicals for CNS depressant activity in many preclinical studies. The assessed activities of Catharanthus roseus leaves could be due to steroids, glycosides and phenolic compounds.
