Abstract

The aim of present work is to formulate and evaluate Anagliptin buccal patch by solvent casting method. Buccal patch has wide importance and it overcomes the limitations of current routes of administration, provides rapid onset of action by releasing drug directly to systemic circulation through oral mucosa by mucoadhesion. The formulated buccal patches were evaluated for various parameters like film thickness, surface pH, folding endurance, weight variation, % moisture loss, ex vivo permeation study, tensile strength, % elongation, drug content uniformity, ex vivo residence time and in vitro dissolution studies. To improve the bioavailability of the drug, buccal patch can be formulated using combination of polymers like HPMC E15, Ethyl cellulose, PVPK90, Glycerine, Methanol, Citric acid, Tween80, Menthol. It may be concluded that the films with HPMC-E15 and Ethyl cellulose (F3) show a convenient residence time and promising drug release, thus seems to be a potential candidate for the development of buccal film for effective therapeutic use. The ex vivo permeation study of the optimized formulation showed a permeation of 86.30% within 8hr which indicate that it would provide an immediate relief due to its faster absorption in oral cavity and is a better alternative to conventional dosage forms. The in vitro drug release of optimized formulation showed drug release of 97.49%. The kinetic data analysis revealed that the formulated films best fit to Higuchi model and follows first order release kinetics. The mechanism of drug release from the film followed non- fickian diffusion.
