Homology modeling and validation of bacterial superoxide dismutase enzyme, an antioxidant

The oxidative stress in humans occurs due to the release of reactive oxygen molecules (superoxide radicals, hydrogen peroxide and hydroxyl radicals) during respiration. The reactive oxygen species damage vital biomolecules of living cells. The enzymes, superoxide dismutases nullify the effect of reactive oxygen molecules. Three types of superoxide dismutases occur in humans' viz., CuZn superoxide dismutase, Mn superoxide dismutase and superoxide dismutase3. The superoxide dismutases can be administered as drugs for the treatment of oxidative stress. In the present paper the amino acid sequence of bacterial Mn superoxide dismutase was retrieved from UniProt server. The amino acid sequence was used as an input data in SWISS MODEL server and Mn superoxide dismutase enzyme three-dimensional model was developed by automated mode. The model quality of Mn superoxide dismutase was checked and validated in PROCHECK server and SPDBV (Swiss PDB Viewer). Further the activity of Mn superoxide dismutase can be improved by modifying its model using sophisticated bioinformatics tools.
