Tannic acid administration ameliorates the levels of hepatic markers, carbohydrate metabolizing enzymes and inflammatory markers in the liver of streptozotocin-induced diabetic albino wistar rats

Tannic acid (TA) is a polyphenol that is found in a variety of plants and fruits, and is well known to have several pharmacological activities. The main goal of the study was to explore the effects of TA on the liver of streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in normal rats by intraperitoneal injection of 50 mg of STZ. The diabetic control rats displayed significantly (p < 0.05) increased hepatic markers such as aspartate transaminase (AST), alanine transaminase (ALT) as well as alkaline phosphatase (ALP) activities, and pro-inflammatory cytokines tumor necrosis factor-alpha (TNF-a) and interleukin-6 (IL-6) activities in the liver. Moreover, the activities of carbohydrate metabolizing enzymes such as glucose-6-phosphatase and fructose-1,6-bisphosphatase was found to be significantly (p < 0.05) increased, while hexokinase, glucose-6-phosphate dehydrogenase and glycogen content significantly (p < 0.05) decreased in the liver of diabetic rats. Diabetic control rat liver histology also displayed several degrees of damages in tissue architecture and vacuoles in the tissue sections. Oral treatment of TA (200 mg/kg bw) to diabetic rats for 45 days impeded the above alterations and were found to be within the normal physiological and metabolic ranges. These findings were also supported by the histological findings, where TA administered diabetic rats showed near normal liver tissue with mild central vein congestion when compared to diabetic control rats. Thus, the present study confirmed that TA has anti-hyperglycemic and anti-inflammatory activities that helped ameliorate adverse effects found in STZ induced diabetic rats.
