Characterization and Optimization of process parameter for maximum Poly (−β-) hydroxyl butyrate production by bacteria isolated from the soil

Poly (ß-hydroxybutyrate) (PHB) is a biodegradable bacterial plastic belongs to the family of polyhydroxyalkanoates (PHA), and it is produced in the stress conditions like excess carbon source and limitation of nitrogen sources. In this study, out of eight isolates, the high amount of PHB granules accumulated isolates was screened by Nile blue stain. Further, the PHB quantified by HPLC, and the functional group was confirmed by FTIR analysis. Peptone was found to be the best nitrogen source which gives 2.01μg/ml of PHB and D-Glucose was found to be the best carbon source which gives 1.44μg/ml of PHB. The maximum yield was obtained when incubated at 35°C (14.0μg/ml) with pH 7 (14.44μg/ml). The present study provides the useful data about the optimized conditions for PHB production, and it can be utilized for the production of biomedical devices, films, porous matrices, microcapsules, microspheres, used in the drμg delivery system, etc.
