Screening of antioxidant and antibacterial activity of methanolic extract of Ipomoea aquatica leaf and stem against bacteria causes skin infection

Bacterial skin infections can be caused by Staphylococcus aureus, Streptococcus pyogenes and Escherichia coli. Currently, bacterial skin infections are treated with antibiotics, which may lead to antibiotic resistance. An alternative option is by obtaining from plant extracts. Ipomoea aquatica were proven to have many pharmacological properties, including antibacterial activity. This study was aimed to screen the antioxidant and antibacterial activity of methanolic extract of I. aquatica leaf and stem against bacteria that causes skin infections. Methanolic extracts [Soxhlet extraction] of I. aquatica leaf and stem were evaluated for total phenolic compound [TPC] and total flavonoid compound [TFC] using gallic acid and quercetin as the control, respectively. Antioxidant activity was tested using DPPH free radical scavenging activity, whereas the antibacterial activity was tested using disc diffusion method. Minimum inhibitory concentration was tested using resazurin as an indicator. All the test was done in triplicate [n = 3, mean ± SD]. Data were analysed using SPSS version 25 [one-way ANOVA]. Flavonoids and phenols were present in both, leaf and stem extract. For TPC, stem [43.18 ± 0.007 mg GAE/g] is higher compared to leaf [20.92 ± 0.007 mg GAE/g], whereas for TFC, leaf [14.88 ± 0.011 mg QE/g] is higher compared to stem [3.00 ± 0.005 mg QE/g]. In DPPH scavenging activity, stem [IC50: 3.84 mg/ml] has higher activity than leaf [IC50: 4.8 mg/ml]. At 80 mg/ml and 100 mg/ml, I. aquatica leaf significantly [P < 0.05] showed higher zone of inhibition compared to stem extract against S. aureus, S. pyogenes and E. coli. However, the zone of inhibition against E. coli is the biggest, followed by S. aureus and then S. pyogenes. Methanolic extract of I. aquatica leaf and stem do have antioxidant and antibacterial activity and can be used to treat bacterial skin infections.
