Antibacterial Screening of Clerodendrum infortunatum leaves: Experimental and Molecular docking studies

Abstract

Over the past decade, herbal and ayurvedic drugs have become a subject of world importance, with both medicinal and economical implications. Clerodendrum infortunatum plants have been used as a source of medicine since ancient time. The major groups of chemical constituents present in the Clerodendrum genus are carbohydrates, phenolics, flavonoids, terpenoids and steroids. Isolated flavonoids like hispidulin, ursolic and cleroflavone and a diterpenoid clerodin possess potent anti-oxidant, anti-microbial, anti-asthmatic, anti-tumor and CNS-binding activities. The present study was conducted to determine the antibacterial potency of Clerodendrum infortunatum leaves with two different solvents ethyl acetate and acetone against Escherichia coli gram negative bacterial strains. The in-vitro screening for antimicrobial activity was carried out by using well-plate techniques. Flavonoids and terpenoids can inhibit the enoyl ACP reductase that present in the Escherichia coli is an essential bacterial enzyme. The enoyl acyl carrier protein reductase of the Escherichia coli species is one of the attractive targets in E. coli associated diseases. By considering the above observations, an attempt for molecular docking studies of flavonoids and terpenoids present in Clerodendrum infortunatum with enoyl ACP reductase (PDB ID: 1C14) by using in silico studies by molinspiration online tool and evaluated for their antimicrobial activity. The compounds showed good activity against Escherichia coli comparable to that of standard drug Ciprofloxacin. Thus, Clerodendrum infortunatum leaves appears to be an effective material for development of antimicrobial drugs. This work explains the evidence-based information regarding the different pharmacological activities with a correlation of chemical constituents available with this plant so that it may serve as a reference for further studies.
