Proteinaceous alpha amylase inhibitor from Macrotyloma uniflorum seeds: In vitro and in vivo studies with Spodoptera litura
Abstract
Alpha amylase inhibitor was purified from the seeds of Macrotyoloma uniflorum by ion exchange and gel filtration chromatography. the inhibitor showed optimum inhibition at ph 7.0 and 40°c. It had a molecular weight of 32360 (± 1023) D and 31416 D as determined by gel filtration and MALDI-toF Ms respectively. It inhibited the alpha amylase from the second instar of Spodoptera litura in a non-competitive manner with a Ki of 0.35 × 10−6 M and LC50 value of 480μg/ml. the effect of the Macrotyoloma uniflorum alpha amylase inhibitor (MuAI) was evaluated by conducting feeding trials on larvae of Spodoptera litura. the MuAI at concentrations of 0.3, 0.5, 0.8 and 1.0% were used for the assay. the maximum average mortality was observed at 1% MUAI concentration.
