Molecular Characterization of Races Fusarium oxysporum f. sp. ciceri Through RAPD Markers

Abstract

The genetic diversity was studied among the 4 races (5 isolates) of Fusarium oxysporum f. sp. ciceri by using 20 RAPD primers of OPF series. Off which 12 primers produced 54 scorable bands with an average of 5 bands per primer. Off 54 bands, 48 bands were polymorphic and level of polymorphism was 81.5 per cent. The primer OPF-6 amplified maximum 8 bands within the size 2402 bp to 468 bp. While primer OPF-14 amplified minimum 2 bands within the size 577 bp to 421 bp respectively. The five RAPD primers OPF-6 (468 bp), OPF-11(1250 bp and 513 bp), OPF-13(1898 bp), OPF-14 (577 bp) and OPF-16 (1089 bp) showed six monomorphic bands, this can be used for development of the SCAR markers for identification of Fusarium oxysporum f.sp. ciceri.The Type Race-4 (Jabalpur and New Delhi) are similar and shows the higher similarity index and grouped under cluster-A. The Type Race-1 (Hyderabad) and Type Race-2 (Kanpur) are grouped under cluster-B. Whereas Type Race-3 (Gurudaspur) shows out-group distinct from other races of Fusarium oxysporum f.sp. ciceri.
