Online published on 3 April, 2013. Abstract Mouth dissolving tablets constitute an innovative dosage form that overcomes the problems of swallowing and provides a quick onset of action. The investigation was undertaken with a view to develop fast dissolving tablets to get the quick onset of action in treatment of suddenly arising allergic reactions like urticaria, pruritis, angeoedema, bronchoconstriction and allergic rhinitis. An attempt has been made to study direct compression for formulation of mouth dissolving tablets of Diphenhydramine HCl. Total Nine formulations using various superdisintegrants namely Sodium starch glycollate, Crospovidone and Croscarmellose sodium were prepared. Taste inhibition has been achieved by using sensory approach. All prepared formulations were evaluated for physico-chemical parameters. The formulations exhibited good disintegration properties with total disintegration time in the range of 20 to 35 sec. along with rapid in vitro drug release of 95% in first 15 minutes, it was concluded that the fast disintegrating/dissolving tablets of Diphenhydramine HCl can be made by direct compression technique using selective superdisintegrants showing quick onset of action followed by pleasant mouth feel, improved patient compliance and effective therapy.
