Abstract In the present study an efficient and successful protocol for enhancement, accumulation and production of wedelolactone from in vitro derived calli and suspension cultures of Eclipta alba was established. The aseptically germinated seedlings were used to raise cultures having different combinations-concentrations of plant growth regulators and precursors. The presence of 6-BA along with other combination of growth regulators had a significant effect on the increase in concentration of wedelolactone and auxin, 2, 4-D (5.0μM) alone had improved wedelolactone (0.018%) content. The mean wedelolactone per cent value was 0.012% leaf of natural plant while the control static MS containing 2, 4-D (5.0μM) + Kin (0.5μM) + 6-BA (0.5μM) had 0.095%. The mean per cent concentration of the wedelolactone in the control suspension culture on MS+2, 4-D (5μM) + Kin (10μM) + 6-BA (1μM) was 0.288% which was about 3.0 fold to the static control cultures. The maximum mean per cent value of wedelolactone was found in phenylalanine (50mg/l) in MS (0.440%) and B5 (0.415%) medium. The mean per cent value of wedelolactone increased to 1.5 fold in phenylalanine from control on MS media. There was 24 fold increases in production of herbal hepatoprotective drug-wedelolactone from E. alba through suspension culture.
