Induction of mammary gland tumour in rats using N-methyl-N-nitroso urea and their histopathology

Abstract

The incidence of Breast cancer among women is increasing day by day in developing countries. It remains a challenge to biologist. Over the past two decade's murine model of mammary cancer provided greater insights into derangements of molecular mechanisms that lead to cancer. Murine models offer a tremendous opportunity to identify such molecular network and their correlation with women breast cancer. These models are used to investigate the factors involved in malignant transformation, invasion and metastasis as well as to examine the response to therapy. Chemical carcinogen induced tumour model has a number of features that make it particularly attractive to study the biology of mammary cancer and for developing and evaluating cancer prevention strategies. N-methyl N- Nitroso Urea (MNU) is in general hormone dependent carcinogenic chemical. In the present investigation N-methyl N-Nitroso Urea (sigma Aldrich, USA) was used for the induction of mammary tumour in female Sprague dawley rats for the study of breast cancer development and histopathological study along with environmental effect on induction of tumour. 35.55% (32/90) of rats were developed mammary tumour after 20 to 25 weeks of first injection of MNU, which is significantly low as reported by other researcher.
