Abstract Background DNA analysis of gynecological swabsin sexualassault casesis a challenge, If we detect a large volume of female material it may obstacle detection of male DNA. The solution of this problem was applying differential DNA extraction, the use of Y-chromosome genetic markers, such as short tandem repeats (STRs), it amplify the low quantities of DNA of male regardless the victim's DNA background, Autosomal DNA STRs were amplified with 28 PCR cycles, whereas 34 PCR cycles were used for Y-STRs, can explain the difference between autosomal DNA STR profiles, and Y-STR profiles Method Y-STRs were amplified with 34 PCR cycles, Therefore, the acquired Y-STR profile is compared only with the Y-STR profile of known suspects, Results In the present case a young male was found dead under the balcony of newly married wife there were a suspect of betrayal, the use of Y-STR used to solve this case, Y-STR can solve the case as the DNA in female vagina belongs to the female boyfriend not the husband, it helps in solving this case Conclusion Y-STR is very useful in solving the cases of sexual assault with a small amount of male DNA.
