Division of Bacteriology and Mycology, Indian Veterinary research Institute, Izatnagar-243122, (Uttar Pradesh). vijendra@ivri.up.nic.in Abstract A study was conducted to evaluate the diagnostic potential of PCR in preserved morbid material infected with Pasteurella multocida responsible for Haemorrhagic septicaemia in cattle and buffaloes. The study also aimed to see the efficacy of different chemical formulations used as tissue preservative in detection of Pasteurella multocida infection in tissue by PCR. The morbid tissue materials from experimentally infected mice with virulent Pasteurella multocida serotype B:2 were preserved in three different preservatives namely phosphate buffered formalin (PBF), Carnoy's solution and Davidson's solution. Pasteurella multocida specific PCR using DNA extracted from morbid materials preserved in all the three preservatives revealed its diagnostic potential with varying degree of success and phosphate buffered formalin (10%) was found the most suitable preservative.
