Abstract Prevalence of MAP was 28.3 and 20.8% by fecal culture and serum ELISA kit, respectively, in three dairy herds located in Mathura district of North India. Herd-wise prevalence in Krishna Balram Gosala, Malwiya Gosala and DDD Farm, Mathura was 20, 30 and 29% and 30, 50 and 17% by fecal culture and ELISA kit, respectively. However, 30.8% cows were positive when fecal samples of 120 cows were screened by direct microscopy. Comparison of fecal culture and ELISA kit by ‘Kappa value’ showed that the two tests had nearly perfect agreement. ‘Indigenous ELISA kit’ used for screening of cows had 50.0 and 90.6% of sensitivity and specificity with respect to fecal culture. Screening of the 34 (28.3% positive) cultures by specific IS900 PCR revealed that 29 (85.2%) samples were positive for MAP. Genotyping of the MAP colonies by IS1311 PCR-REA showed the presence of ‘Bison type’ genotype in dairy herds of Mathura city. Using ELISA kit for screening of 224 serum samples from 5 dairy herds in Mathura district, 23.6% cows were in strong positive category and were considered positive. ‘Herd-wise’ prevalence of JD was 36.3, 35.0, 16.0, 29.0 and 33.3% in Krishna Balram Goshala, Malwiya Goshala, DDD Farm, Mathura, Panchayati Goshala and Veterinary Clinics, respectively. ‘Bison type’ genotype has also been reported previously from goats and sheep in Mathura district. These reports indicated that ‘Bison type’ genotype was predominant genotype in Mathura district of the country and was shared by three important domestic livestock species. Top |
