Biochemical and ultra-structural hepatic perturbations and its recovery in fresh water fish- Channa punctatus (Bloch.) exposed to sub-lethal concentration of nigrocine black (a tannery dye)

The aim of this investigation was to compare biochemical and ultra-strucutral perturbations induced by nigrocine black (NB) in the liver of fresh water fish- Channa punctatus (Bloch.) exposed for 56 days to sub-lethal dose of NB (1/10th of LC50 96h) and then replaced for further 56 days in NB-impoverished water (normal water). Toxicity of NB was clearly demonstrated at various days of exposure (11, 22, 33, 44 and 56). Activity of both the transaminase (SGOT and SGPT) were increased significantly (P < 0.05) throughout the exposure period. Activity of SALP also increased significantly up to the middle of exposure period but decreased thereafter till the end. Hepatocytes of exposed fishes showed ctyoplasmic vacuolations, accumulation of lipid droplets and heterochromatin. Perichromatin granules were increased and progressively formed small clusters. Glycogen rosettes enclosed by membrane and increasing number of secondary lysosomes were found. Furthermore, few hepatocytes appeared with macrophages and residual body. During recovery period, the fishes showed significant recovery in SGPT and SALP activities throughout the recovery period with maximum at the end but they did not show recovery in SGOT activity. Similarly, the hepatocytes were also observed to be normal in many respect when compared with the hepatocytes of control fish.
