Production of ecofriendly silver nanoparticle and evaluation of its potential antimicrobial activity

Abstract

Fish scales are a major by-product of the fish-processing industry, causing wastage and pollution. The waste management and converting it into useful resources in an ecofriendly way is challenging one. Nanotechnology is a field that is burgeoning day by day, making an impact in all walks of human life. By applying this technology, silver nano particles were produced from the collagen extracted from the fish waste with cold 0.5M acetic acid and purified by alternative precipitation with sodium chloride and dialysis against 0.02M disodium hydrogen phosphate. The purified collagen was characterized with the help of SDS-PAGE. 10ml of collagenous substance was added to the 90ml of aqueous solution of 1Mm of silver nitrate and 0.5M of SDS and 0.5M of tri sodium citrate was added along with that and keep at room temperature for 5hrs. The pellets were collected to obtain 1g of wet sample and resuspended in 10ml of 1Mm Silver nitrate solution and incubated for 24hours at 3000rpm followed by sonication and centrifugation. The filtrate sample without AgNo3 act as a control, periodically aliquots of the reaction mixture were taken out and the absorbtion was measured in a Hitachi UV-VIS spectrophotometer. The samples were analyzed through FTIR. Finally the antibacterial activities of synthesisied silver nanoparticles were done on human pathogenic Escherichia coli and Pseudomonas aueroginosa by standard disc diffusion method.
