Formulation and Evaluation of Loratadine Fast Dissolving Films

The object of the study was to prepare Fast dissolving Films (FDF) of Loratadine which can be useful in the symptomatic relief of allergy such has hay fever (allergic rhinitis), urticaria (hives) and other skin allergies. Fast dissolving Films were prepared by solvent casting method using various polymers such as HPMC E15, PVP and Sodium Alginate alone and in combinations along with PEG 400 as a plasticizer and Tween 80 as surfactant. Prepared Loratadine films were evaluated for film forming capacity, visual appearance, weight uniformity, thickness, drug content uniformity, folding endurance, disintegration time, in-vitro loratadine release, and stability studies. Among all the polymers employed sodium alginate and PVP (3:1 ratio) showed desired film forming capacity. Combination of polymers resulted in least disintegration time. A film formed of F9 formulation was transparent, smooth and homogenous with best film forming property among all the film forming agents. Films in formulation F9 showed its in vitro disintegration time was 39 seconds and its cumulative percent drug release was 100% within 14 min which is remarkable in comparison to other formulations. FTIR and stability studies of formulation F9 revealed the stability of the formulation.
