Evaluation of the Antioxidant and Potential binding affinity of Cyamopsis tetragonoloba seed against the receptor responsible for Gouty Arthritis

Abstract

Guar gum is an herbal drug that is used for treating the diabetes and cancer which is also used as a dietary supplement. It has properties as anti-microbial, anti-tumor, and anti-inflammatory. Hyperuricemia or Gout is an acute inflammation that can be found in joints with the indications of redness and swollen knees. The aim of our study is to know the potential effect of Cyamopsis tetragonoloba through in vitro and in silico analysis. The antioxidant activities like catalase, peroxidase, DPPH assay and total phenolic content were evaluated in the aqueous, ethanolic and methanolic extract of Cyamopsis tetragonoloba seed. Ligands of Cyamopsis tetragonoloba seed were obtained from the literature and the 3d structure of the ligands were retrieved from Pubchem online server followed by Corina molecular network. Receptors responsible for causing gouty arthritis were obtained from RCSB protein data bank in the format of 3d structure. Patch dock online serve was used to perform the docking analysis which was then visualized and analyzed in PyMOL. Extract of Cyamopsis tetragonoloba seed shown good antioxidant potential in which aqueous extract was observed to show better antioxidant activity. Docked complex with Hexopyranosyl hexopyranoside has shown better interaction with higher binding residues than the other ligands. Further, in vivo studies can be carried out to known the potential activity of this ligand against gouty arthritis.
