Effect of Linagliptin and Niclosamide on Streptozotocin Induced Diabetic Neuropathy in Rats

Abstract

Diabetic neuropathy is common microvascular complication of diabetes mellitus. Objective of research was to evaluate the effect of Linagliptin and Niclosamide on Streptozotocin (STZ) induced neuropathy in experimental diabetic Wistar Rats. Here, animals were fed with high fat diet for about two weeks after that injected with streptozotocin 30 mg/kg i.p. dual dose in consequent week for production of diabetes. Animals were separated into eight groups. Locomotor activity, Tail immersion, hot plate and motor coordination after two week of diabetes induction were used for assessed Diabetic neuropathy. Disease animals treated with Linagliptin (3, 5mg/kg p.o) and Niclosamide (30, 75mg/kg i.p.) and their combination as Linagliptin + Niclosamide (5, 75mg/kg). Pregabalin(10 mg/kg i.p.) given as standard to one group. At the end of treatment to measure serum glucose, cholesterol, triglyceride level blood sample was taken. Sciatic nerve was removed and investigates for level of antioxidant such as SOD, CAT, LPO, NO etc. In order to assess role of Linagliptin and Niclosamide in the relevant activity, further histopathological studies provide a supportive evidence for this study to show protective effect of drugs. Treatment with Linagliptin and Niclosamide gives protective effect in neuropathy associated with streptozotocin induced diabetes in rats. Their combination for four week showed significant fall in serum glucose, cholesterol, triglyceride level and also improve SOD, CAT level while significant fall in LPO and NO level in sciatic nerve. Further combination showed marked improvement. Thus, combination of these drugs could be better in ameliorating diabetic neuropathy in STZ induced diabetes.
