Abstract The present study was conducted to develop an efficient protocol for rapid in vitro micropropagation by multiple shoot induction from nodal segment of Cissus quadrangularis, an important medicinal plant, on MS-medium supplemented with different concentration and combinations of kinetin, 2, 4-dichlorophenoxyacetic acid, 6- benzylaminopurine and zeatin. Four multiple shoots were observed in MS medium supplemented with zeatin (1 mg L−1). In some cases, 2, 4-dichlorophenoxyacetic acid (1 mg L−1) also showed better results. The maximum mean length of the shoots was recorded on MS medium fortified with 6-benzylaminopurine (2 mg L−1). In some cases 2, 4-dichlorophenoxyacetic acid (1 mg L−1) and 6-benzylaminopurine (6 mg L−1) in combination with kinetin (1 mg L−1) also showed good results. The regenerated shoots were rooted better on MS-basal medium (80%) without plant growth regulators as compared to MS medium with indole-3-butyric acid (1 mg L−1). This method may successfully be employed for large scale multiplication and in vitro conservation of C. quadrangularis. Top Keywords Cissus quadrangularis, medicinal plant, micropropagation. Top | |
