To know the dynamic changes in immune-effector activities of Indian major carp, rohu (Labeo rohita) against Aeromonas hydrophila, experimental Motile Aeromonas Septicemia (MAS) was developed in rohu using intraperitoneal injection (0.2ml/fish) with live A. hydrophila at a concentration of 1×106 cfu/ml. Dynamic changes in humoral and cellular immune responses were measured at 5th and 15th day post inoculation (PI). A three tier ELISA, involving an anti-fish enzyme immunoconjugate was developed for the measurement of antibody titre. A. hydrophila sensitized rohu showed enhanced innate immune-effector activities viz. respiratory burst activity and nitric oxide production, in 5th day PI period. Specific humoral and cellular immune responses, viz. antibody production and lymphoproliferation assay, respectively, were gradually increased in latter phase (15th day PI) of sensitization. Theses basic information should be exploited meticulously during immunodiagnostic and/or immunoprophylactic preparations against MAS in rohu.
